Intein-Mediated Protein Ligation Of Expressed Proteins

View page
US Patent:
20050100987, May 12, 2005
Filed:
Oct 7, 2004
Appl. No.:
10/960905
Inventors:
Thomas Evans - Somerville MA, US
Ming Xu - Hamilton MA, US
Shaorong Chong - Somerville MA, US
International Classification:
C12P021/06
C07H021/04
C12N009/00
C12N001/18
C12N005/06
C12N015/74
US Classification:
435069100, 435320100, 435471000, 435325000, 435348000, 435254200, 435252100, 435183000, 530350000, 536023200
Abstract:
A method for the ligation of expressed proteins which utilizes inteins, for example the RIR1 intein from , is provided. Constructs of the Mth RIR1 intein in which either the C-terminal asparagine or N-terminal cysteine of the intein are replaced with alanine enable the facile isolation of a protein with a specified N-terminal, for example, cysteine for use in the fusion of two or more expressed proteins. The method involves the steps of generating a C-terminal thioester-tagged target protein and a second target protein having a specified N-terminal via inteins, such as the modified Mth RIR1 intein, and ligating these proteins. A similar method for producing a cyclic or polymerized protein is provided. Modified inteins engineered to cleave at their C-terminus or N-terminus, respectively, and DNA and plasmids encoding these modified inteins are also provided.

Metals For Razor Blade Applications

View page
US Patent:
20220347876, Nov 3, 2022
Filed:
May 3, 2021
Appl. No.:
17/306685
Inventors:
- Boston MA, US
Ming Laura Xu - Natick MA, US
International Classification:
B26B 21/60
B05D 1/02
Abstract:
Razor blades with two or more materials and/or methods of fabricating the same. In one example, a bimetal razor blade can comprise a blade body and a blade edge. The blade body can have a length and a width. The blade body can be formed of a first substrate portion comprising a first material having a first hardness. The blade edge can extend along the length. The blade edge can be formed of a second substrate portion coupled to the first substrate portion. The second substrate portion can comprise a second material having a second hardness that is distinct from the first hardness. The first and second materials can be metals.

A61K 31/506
A61K 31/353
A01K 67/027
A61P 39/00
Abstract:
This document provides methods and materials for treating aging and/or improving transplant outcomes. For example, methods and materials for using one or more senotherapeutic agents to reduce risk of transplant rejection are provided. Non-human animal models for transplant rejection as well as methods for using such non-human animal models to identify agents having the ability to reduce transplant rejection are provided as are non-human animal models for aging and methods for using such non-human animal models to identify agents having the ability to treat aging or the ability to slow the effects of aging.

Bent Razor Blades And Manufacturing Of Such Razor Blades

View page
US Patent:
20180126574, May 10, 2018
Filed:
Oct 16, 2017
Appl. No.:
15/784453
Inventors:
- Boston MA, US
Ming Laura Xu - Natick MA, US
John Lawrence Maziarz - Framingham MA, US
International Classification:
B26B 21/56
B26B 21/22
B26B 21/06
B26B 21/40
Abstract:
A method of manufacturing a razor blade by providing a razor blank strip having an edge. The razor blank strip is hardened to a hardness greater than 600HV. A pre-bending zone that is spaced apart from an edge of the razor blank is melted with a laser beam. The pre-bending zone is cooled after melting to form a re-solidification portion. The razor blank strip is segmented into individual blades. The individual blades are bent in the pre-bending zone.

US Patent:
20170136640, May 18, 2017
Filed:
Nov 13, 2015
Appl. No.:
14/940858
Inventors:
- Boston MA, US
Ming Laura Xu - Natick MA, US
Yongqing Ju - Medfield MA, US
Bin Shen - Allston MA, US
Matthew Robert Stone - Oxford MA, US
International Classification:
B26B 21/60
B26B 21/56
Abstract:
A razor blade having a substrate with a cutting edge being defined by a sharpened tip is provided where the substrate is comprised of a molybdenum (Mo) content of about 1.6% to about 5% by weight of composition and thicknesses of 1.60-1.75 μm and 9.25-10.00 μm measured at a distance of 4 and 40 μm from the blade tip, respectively. This thickness ratio is between 0.165-0.185. The substrate thickness is about 2.70-3.00 μm at 8 μm from the blade tip, about 4.44-5.00 μm at 16 μm from the blade tip with a thickness ratio measured at 4 μm and 8 μm between 0.56-0.62, and a thickness ratio measured at 4 μm and 16 μm between 0.32-0.40. The edge shape may be defined by equation w=adwith “a” between 0.50-0.62 and “n” between 0.76-0.80. A semi-included angle less than 7 is measured at a distance of 40 μm or greater from the blade tip. This substrate will have substantially no cracks.
