Nucleic Acid Melting Analysis With Saturation Dyes

View page
US Patent:
7456281, Nov 25, 2008
Filed:
Jul 13, 2006
Appl. No.:
11/485851
Inventors:
Virginie E. Dujols - Sandy UT, US
Assignee:
Idaho Technology, Inc. - Salt Lake City
International Classification:
C07D 239/00
C07D 239/02
C07D 215/00
US Classification:
544242, 544245, 544334, 548152
Abstract:
Methods are provided for nucleic acid analysis wherein a target nucleic acid is mixed with a dsDNA binding dye to form a mixture. Optionally, an unlabeled probe is included in the mixture. A melting curve is generated for the target nucleic acid by measuring fluorescence from the dsDNA binding dye as the mixture is heated. Dyes for use in nucleic acid analysis and methods for making dyes are also provided.

Jun 4, 2007
Appl. No.:
11/757836
Inventors:
Carl T. Wittwer - Salt Lake City UT, US
C. Wade Dummer - Salt Lake City UT, US
Virginie E. Dujols - Salt Lake City UT, US
Assignee:
University of Utah Research Foundation - Salt Lake City UT
International Classification:
C12Q 1/68
C07H 21/02
C07H 21/04
US Classification:
435 6, 536 231, 536 243
Abstract:
Methods of identifying the type of a cell are provided, the methods comprising determining the Tprofile of a sample rRNA or fragment thereof from the cell using a double stranded nucleic acid-specific dye, wherein a match between the determined Tprofile and the Tprofile of a corresponding rRNA or fragment thereof of a cell from a known cell type indicates that the sample rRNA is from the known cell type.

US Patent:
7582429, Sep 1, 2009
Filed:
Oct 22, 2003
Appl. No.:
10/531966
Inventors:
Carl T. Wittwer - Salt Lake City UT, US
Gudrun Reed - Salt Lake City UT, US
Virginie Dujols - Sandy UT, US
Luming Zhou - Salt Lake City UT, US
Assignee:
University of Utah Research Foundation - Salt Lake City UT
International Classification:

C12P 19/34
C07H 21/04
US Classification:
435 6, 435 912, 536 2433
Abstract:
Methods are provided for nucleic acid analysis wherein a target nucleic acid that is at least partially double stranded is mixed with a dsDNA binding dye having a percent saturation of at least 50% to form a mixture. In one embodiment, the nucleic acid is amplified in the presence of the dsDNA binding dye, and in another embodiment a melting curve is generated for the target nucleic acid by measuring fluorescence from the dsDNA binding dye as the mixture is heated. Dyes for use in nucleic acid analysis and methods for making dyes are also provided.
