Molecular biology: Designed and developed experimental plans for reagents, system integration for nucleic acids based assays including Recombinant DNA technology, cloning, DNA sequencing, site-directed mutagenesis, screening of genomic and cDNA libraries, southern blotting, Telomerase Terminal restriction Fragment analysis (TRFs), RNA, RT-PCR, multiplex PCR, real time qPCR, Northerns, siRNA/shRNA, microRNAs, and Gene expression profiling. Product development and high throughput automation. Biochemical assays: ECL and LI-COR based quantitative immunoblots, immunoprecipitations - Recombinant protein expression/purification/charac... affinity chromatography, Thin Layer chromatography - Protein-protein interactions: Yeast/Mammalian two hybrid screen, binding assays (in vivo as well as in vitro) - Protein-nucleic acid interaction: Electrophoretic gel mobility shift assays, co-precipitations. Microscopy (bright field, fluorescence including confocal, laser capture microscope), IHC, Immunocytochemistry, high content imaging and analysis. In vivo Pharmacology models: Small animal experimental techniques and toxicology studies including genotyping, surgical procedures, and preparation of MEFs. Cell biology: Extensive cell culture experience including primary cells (2D, 3D matrigel), transient and stable transfections, retro- and adenoviral mediated cell transfection, stable cell line development, isolation & characterization of primary tumor cells, mobility assays, high-throughput format assays using high content imaging, optimization in 96-well and 384-well formats, MTT Cell Proliferation Assay, BrdU-PI staining, flat cell assays, colony formation assays, immunocytochemistry, ELISA, apoptosis, viability, and flow cytometry.
