Abstract:
The invention provides a flow system and method for reliable multiparameter data acquisition and particle sorting. In accordance with the invention, a flow system assesses changes in the pattern of data collected in successive time intervals and actuates one or more corrective actions whenever the changes exceed predetermined limits. The present invention overcomes problems associated with collecting data and sorting and enumerating particles in flow cytometry systems that operate for prolonged periods or that must accommodate samples that vary widely in quality.

Abstract:
A cartridge, in particular a disposable cartridge, for use in an electrowetting sample processing system. The cartridge contains an internal gap with at least one hydrophobic surface for enabling an electrowetting induced movement of a microfluidic droplet that has magnetic beads and further has a bead accumulation zone, into which the microfluidic droplet is transferable by electrowetting force and the magnetic beads are exposable to a magnetic force of a bead manipulation magnet. The internal gap has a bead extraction opening adjacent to the bead accumulation zone. The bead extraction opening provides a passage from the gap to an exterior space of the cartridge and is configured to removably receive the bead manipulation magnet for enabling an extraction of the magnetic beads from the microfluidic droplet by a removal of the bead manipulation magnet.

Methods and systems are disclosed for controlling a particle analyzer based at least in part on a generated entrainment factor for a sample. The features include flowing a sample with a series of particles through the particle analyzer, detecting events and calculating an expected frequency of those events based on a distribution, such as a Poisson distribution, and measuring an observed frequency of particle events. An entrainment factor may be generated from a ratio of observed event frequency to expected event frequency. Further adjustment to the particle analyzer maybe performed based on the indicated entrainment factor such as adjusted sorting bias.

Abstract:
Methods and systems are disclosed for generating an entrainment factor in a flow cytometry sample.
