Abstract:
Translation systems and other compositions including orthogonal aminoacyl tRNA-synthetases that preferentially charge an orthogonal tRNA with an iodinated or brominated amino acid are provided. Nucleic acids encoding such synthetases are also described, as are methods and kits for producing proteins including heavy atom-containing amino acids, e. g. , brominated or iodinated amino acids. Methods of determining the structure of a protein, e. g. , a protein into which a heavy atom has been site-specifically incorporated through use of an orthogonal tRNA/aminoacyl tRNA-synthetase pair, are also described.

Compositions and methods of producing components of protein biosynthetic machinery that include orthogonal leucyl-tRNAs, orthogonal leucyl-aminoacyl-tRNA synthetases, and orthogonal pairs of leucyl-tRNAs/synthetases, which incorporate photoregulated amino acids, OMe-L-tyrosine, α-aminocaprylic acid, or o-nitrobenzyl cysteine into proteins are proteins are provided in response to an amber selector codon. A clustered storage system includes a plurality of storage devices, each of which contributes a portion of its memory to form a global cache of the clustered storage system that is accessible by the plurality of storage devices. Cache metadata for accessing the global cache may be organized in a multi-layered structure. In one embodiment, multi-layered structure has a first layer first including a first address array, and the first address array include addresses pointing to a plurality of second address arrays in a second layer. Each second address array in the second layer includes addresses, each of which points to data that has been cached in the global cache.

Abstract:
An embodiment method creates, at a first data center, a first storage volume for an application that is started at the first data center, communicates with a second data center for creating a second storage volume for the application at the second data center to replicate the first storage volume, and sends information identifying the application to the second data center. The second data center creates the second storage volume, and establishes a mapping between the application and the second storage volume using the information identifying the application, where the second data center identifies the second storage volume for the application using the mapping when the application is migrated from the first data center to the second data center.
