Abstract:
Vaccines and methods for inducing an immune response in a ruminant. The vaccine composition including pathogen and tick-derived antigens and a carrier or diluent. The method for inducing an immune response in a ruminate to provide immune protection which reduces the infection of ticks by and/or prevents the transmission of the pathogen includes the steps of administering to the ruminant an effective amount of the vaccine composition having at least one antigen member of the group comprising at least one antigen member of the group comprising (i) recombinant MSP1a surface protein antigen of , (ii) a subunit of recombinant MSP1a surface protein antigen of and (iii) recombinant MSP1a surface protein antigen or subunits thereof in combination with antigen preparation derived from infected cultured tick IDE8 cells and/or other pathogen and tick-derived antigens, and a carrier or diluent.

Abstract:
The invention is directed to methods of culturing rickettsiae in Ixodes scapularis cell lines. The methods of the invention provide for culture of microorganisms such as Anaplasma marginale, Ehrlichia canis, and Rickettsia rickettsii. A method of the invention involves incubating a rickettsia with an Ixodes scapularis tick cell culture in a culture medium under reduced oxygen and increased CO. sub. 2 at a sufficient temperature until growth of the rickettsia is detected. The culture medium comprises a medium suitable for the growth of invertebrate cells supplemented with an organic buffer. The cell culture method can be used in large scale production of rickettsia containing products useful in diagnostic assays and vaccine preparations.
