The inventions provide microscopes for imaging samples within wells of multi-well plates. Microscopes of the disclosure include a beam homogenizer system that shapes a beam from a light source into a shape specific to the bottom of a well of a multi-well plate. In particular, microscopes of the disclosure can illuminate wells for imaging by passing light through a prism that is beneath the sample. The light enters the prism from the side and as refracted into the well at a steep angle such that the light only illuminates about a bottom ten microns of the well. The beam homogenizer shapes the light from the light source so that, instead of hitting the prism as a spot with an irregular shape, the light enters the prism in a substantially rectangular pattern with homogeneous optical power level over the pattern.

Database replication is performed by tracking sequence numbers associated with binary change logs. At a producer cluster, a binary change log is generated based on a write transaction made to a database partition in the producer cluster. Included with the binary change log is a sequence number corresponding to the write transaction. The binary change log is transmitted from the producer cluster to a consumer cluster, where a determination is made whether to apply the binary change log based on the included sequence number. The binary change log is then applied to a database partition in the consumer cluster. The sequence number is stored at the consumer cluster by combining the sequence number with a numerical range having a first number and a second number, where the second number has a value corresponding to the most recent binary change log applied to the database partition in the consumer cluster.

A system and method for correction of aberrations in a solid immersion microscopy system using a deformable mirror. A solid immersion lens is provided having a surface configured to make optical contact with a nearly planar surface of a substrate, an object to be imaged disposed on the opposite side of the substrate. A convex surface of the solid immersion lens faces an objective lens. A deformable mirror assembly, including a plurality of individually controllable actuators, receives light transmitted from the object. A control system controls in communication with the deformable mirror assembly provides individual actuation of each of the actuators of the deformable mirror to compensate or counteract the effects of aberrations.
